【摘要】 目的 通過(guò)觀察姜黃素對(duì)哮喘大鼠肺泡灌洗液總細(xì)胞數(shù)及嗜酸性粒細(xì)胞(Eos)計(jì)數(shù)、肺組織粒細(xì)胞-巨噬細(xì)胞集落刺激因子(GM-CSF)表達(dá)的影響,探討姜黃素對(duì)哮喘的治療作用。方法 36只雄性SD大鼠隨機(jī)分為正常對(duì)照組(A組)、哮喘模型組(B組)、姜黃素治療組(C組)各12只,采用卵蛋白(OVA)致敏大鼠哮喘模型,觀察三組動(dòng)物哮喘發(fā)作癥狀、肺泡灌洗液總細(xì)胞數(shù)及嗜酸性粒細(xì)胞計(jì)數(shù)情況及原位雜交方法檢測(cè)肺組織中GM-CSFmRNA含量變化。結(jié)果 B組大鼠哮喘發(fā)作癥狀最明顯,C組大鼠癥狀輕,A組無(wú)癥狀。B組肺泡灌洗液總細(xì)胞數(shù)及嗜酸性粒細(xì)胞計(jì)數(shù)均明顯高于其余兩組; C組明顯少于B組; A組最少。肺組織中GM-CSFmRNA表達(dá)A組微弱表達(dá), C組較強(qiáng), B組表達(dá)最強(qiáng)。結(jié)論 姜黃素可抑制哮喘大鼠的氣道炎癥,其作用機(jī)制可能是通過(guò)抑制GM-CSF的表達(dá)而導(dǎo)致嗜酸性粒細(xì)胞生成減少及存活時(shí)間縮短來(lái)實(shí)現(xiàn)。

【Abstract】 Objective To observe the effects of curcumine on granulocyte macrophage colony stimulating factor expression and airway inflammation in asthmatic rats. Methods 36 male Sprague-Dawley rats were randomly divided into normal control(A group), model group(B group),curcumine group(C group), 12 rats in each group. Rats were sensitized and challenged continually with ovalbumin(OVA) for the purpose of induction of asthma in rats. To observe asthmatic attack and airway inflammation in asthmatic rats. Granulocyte macrophage colony stimulating factor in rat pulmonary tissues was detected by using in situ hybridization method. Results Asthmatic attack obviously in B group,gently in C group, Absence of symptoms in A group. The total cell amount in bronchoalveolar ravage fluid and eosin Phil were obviously higher in B group than C group and A group. The expression of GM-CSFmRNA in C group was significantly higher than that in A group and it was lower than that in B group.Conclusions The effect of curcumine on airway inflammation in asthmatic rats may be correlated with its inhibition on over expression of GM-CSF in lung tissue.醫(yī).學(xué)全.在.線網(wǎng)站quanxiangyun.cn

支氣管哮喘是一種特殊的氣道炎癥,主要表現(xiàn)為氣道高反應(yīng)與變異性較大的可逆性氣流阻塞。有研究表明[1],粒細(xì)胞-巨噬細(xì)胞集落刺激因子(GM-CSF)在炎癥過(guò)程中起核心作用。姜黃素因其在抗炎、抗纖維化、抗腫瘤、抗氧化等作用而倍受學(xué)者們的關(guān)注[2]。本研究通過(guò)觀察姜黃素對(duì)哮喘大鼠氣道炎癥、肺組織粒細(xì)胞-巨噬細(xì)胞集落刺激因子(GM-CSF)表達(dá)的影響,探討姜黃素對(duì)哮喘的治療作用。
