【摘要】 目的:在卵巢癌腹腔移植瘤模型中進行卵巢癌血管生成擬態(tài)的形態(tài)學(xué)觀察并檢測基質(zhì)金屬蛋白酶2(MMP2)的表達(dá)情況。方法:卵巢癌細(xì)胞SKOV3接種裸鼠腹腔,建立卵巢癌腹腔移植瘤模型,在同一張切片上以抗鼠CD34標(biāo)記鼠血管內(nèi)皮細(xì)胞,PAS染色行基底膜樣結(jié)構(gòu)標(biāo)記。并應(yīng)用免疫組化和RT-PCR方法比較MMP2在卵巢癌腹腔移植瘤中央和邊緣區(qū)域組織的表達(dá)差異。結(jié)果:卵巢癌腹腔移植瘤中央?yún)^(qū)域可見卵巢癌細(xì)胞圍成管狀結(jié)構(gòu),中間可見紅細(xì)胞,未見CD34陽性細(xì)胞出現(xiàn), PAS陽性物質(zhì)呈顆粒狀彌散分布在卵巢癌細(xì)胞漿內(nèi),有的貼附在管腔內(nèi)側(cè)。免疫組化提示MMP2蛋白陽性表達(dá)率在腫瘤中央?yún)^(qū)域組織比腫瘤邊緣區(qū)域明顯增高,RT-PCR分析顯示腫瘤組織中央?yún)^(qū)域MMP2表達(dá)與邊緣區(qū)域相比有顯著性差異。結(jié)論:卵巢癌細(xì)胞分泌PAS陽性物質(zhì),可能參與構(gòu)建腫瘤的血管基質(zhì)重塑。卵巢癌中央?yún)^(qū)域可見血管生成擬態(tài),腫瘤中央?yún)^(qū)域MMP2表達(dá)高于腫瘤邊緣區(qū)域,MMP2表達(dá)是否與卵巢癌血管生成擬態(tài)形成有關(guān),仍需今后進一步研究。

Morphologic observation of vasculogenic mimicry and expression of MMP2 in ovarian carcinoma

[Abstract] Objective: To determine the formation of vasculogenic mimicry and the expression of matrix metalloproteinase 2(MMP2) in ovarian carcinoma in vivo, after the transplantable tumor model of nude mice was established. Methods: Ovarian carcinoma cells SKOV3 were implanted into peritoneal cavity of nude mice. When the transplantable tumor developed, sections of paraffin-embedded and formaldehyde-fixed tissues were stained with PAS and CD34 to study the morphology and distribution of vasculogenic mimicry. Expression of MMP2 was detected by immunohistochemical staining and RT-PCR in the center and the marginal part of the tumor. Results: Vasculogenic mimicry, when PAS was positive but CD34 was negative, appeared in the center of tumor tissues. Ovarian carcinomatous cells formed patterned network with red blood cells in it. PAS positive substance was distributed over the cytoplasm of ovarian carcinomatous cells and sometimes adhere to the wall of the network. The levels of MMP2 mRNA and protein were higher in the center of the tumor tissue than those in the marginal area. Conclusion: Ovarian carcinomatous cells secrete PAS positive substances, which is related to remodeling vascular matrix in tumor. Vasculogenic mimicry appears in the center of tumor tissues. Expression level of MMP2 in the center of tumor tissues is higher than that in the marginal area. It must be further studied whether MMP2 takes part in formation of vasculogenic mimicry.

葡萄膜黑色素瘤、乳腺癌、前列腺癌肝癌、軟組織腫瘤以及卵巢癌等高度惡性腫瘤中存在血管生成擬態(tài)[1-3]。這些惡性腫瘤細(xì)胞在沒有血管內(nèi)皮細(xì)胞的參與下直接形成特有的微循環(huán)管道,有學(xué)者報道通過CD31和PAS雙重染色來觀察惡性黑色素瘤血管生成擬態(tài)的形態(tài)學(xué)特征[4]。本實驗建立裸鼠卵巢癌腹腔移植瘤模型,采用抗鼠CD34標(biāo)記鼠血管內(nèi)皮細(xì)胞,PAS染色行基底膜樣結(jié)構(gòu)標(biāo)記來進行體內(nèi)卵巢癌血管生成擬態(tài)的形態(tài)學(xué)觀察和定位,并應(yīng)用免疫組化和RT-PCR法比較基質(zhì)金屬蛋白酶2(matrix metalloproteinase 2,MMP2)在卵巢癌腹腔移植瘤中央和邊緣區(qū)域組織的表達(dá)差異。
